Wild-type EGF is a potent mitogen for cells of epithelial, mesenchymal and neural origin, driving proliferation, migration and survival, and is one of the most widely used growth factors in cell culture, serving as a core component of media for keratinocytes, epithelial cells, organoids, neural stem cells and induced pluripotent stem cells. It is generated by proteolytic release of a 53-residue domain from a large membrane-anchored precursor, pro-epidermal growth factor, and adopts a compact two-subdomain fold built around three conserved disulfide bonds and a central antiparallel beta-sheet, the architecture that defines the EGF-like domain family.
Signaling upregulates transcriptional programs governing (i) cell cycle entry and proliferation via MYC, FOS, JUN and cyclin D1, (ii) survival via PI3K/AKT and BCL-2 family members, (iii) epithelial migration and wound repair via RAC1 and cytoskeletal remodelling, and (iv) STAT-dependent transcription. Wild-type EGF is expressed at high levels in salivary gland, kidney and pancreas, and is abundant in saliva, urine, milk and plasma.
